Makwassie (Makwasse or Maquassi to some) is a small town in the North West Province of South Africa, just 14 km from Wolmeransstad, on the R505 between Wesselsbron in the Free State Province and Wolmeransstad in the North West.
The name Makwassie is a corruption of the San/Bushman name "Maquassi" for an aromatic wild spearmint.
Farming is the main economic activity in the Makwassie area, where maize, sorghum, groundnuts and milk is produced.
Makwassie was the site of the first christian mission station north of the Vaal River, established in 1822 by Wesleyans Samuel Broadbent and Thomas Hogson. Samuel Broadbent documented the building of the Mission house and the quick establishment of the town today known as Makwassie.

The Makwassie Town Hall was built in 1910.
The Makwassierante Conservation Area offers hiking trails through this rugged conservancy, which hosts a number of game species.
